چکیده

Norcantharidin (NCTD) is a demethylated analogue of cantharidin which is an active ingredient of Chinese medicine-Mylabris. It has been used as an anticancer drug for the treatment of colon cancer, primary hepatoma, carcinomas of esophagus and breast cancer, leukopenia in China for many years (Wang, 1989; Fang et al., 1993). Previous studies have shown that NCTD could suppress the invasion and metastasis of colorectal adenocarcinoma CT26 cells in vitro and in vivo models. In addition, NCTD could mediate a Fas-dependent apoptotic cell death in colon cancer cells such as CT-26, HT-29 (Chen et al., 2008; Peng et al., 2009). However, there is an unexpected discovery that micro-vessels was less in the group treated with NCTD than the control group when we studied the effect of NCTD on HCT116 cell xenografts in nude mice. Hence, we suppose whether NCTD could inhibit angiogenesis of human colorectal cancer, which maybe a new mechanism for anticancer effect of NCTD. Angiogenesis is a hallmark of tumor in order to supply the tumor with its metabolic requirements (Hanahan and Weinberg, 2000; Heath and Bicknell, 2009). Mechanistically, angiogenesis is a proliferation of blood vessel networks from the pre-existing vasculature (Pang and Poon, 2006) and penetrates into cancerous tissue, plays an essential role in carcinogenesis, cancer

In vitro combination therapy of pathologic angiogenesis using anti-vascular endothelial growth factor and anti-neuropilin-1 nanobodies

Objective(s): Emergence of resistant tumor cells to the current therapeutics is the main hindrance in cancer treatment. Combination therapy, which mixes two or more drugs, is a way to overcome resistant problems of cancer cells to current treatments.  Nanobodies are promising tools in cancer therapy due to their high affinity as well as high penetration to tumor sites....

In vivo immunotherapy of lung cancer using cross-species reactive vascular endothelial growth factor nanobodies

Objective(s): Lung cancer is the main leading cause of cancer death worldwide. Angiogenesis is the main step in proliferation and spreading of tumor cells. Targeting vascular endothelial growth factor (VEGF) is an effective approach for inhibition of cancer angiogenesis. Nanobodies (NBs) are a novel class of antibodies derived from the camel. Unique characteristics of Nbs like their small size ...

Aerial Parts of Peucedanum chenur Have Anti-Cancer Properties through the Induction of Apoptosis and Inhibition of Invasion in Human Colorectal Cancer Cells

Background: The Peucedanum species have many pharmacological effects due to the presence of coumarins, flavonoids, phenolic compounds, and essential fatty acids in these species. In this study, for the first time, the anticancer activity of Peucedanum chenur methanolic extract via the induction of apoptosis and inhibition of invasion in HCT-116 human colon cancer cells was investigated. Methods...

Assessment of Antitumor Activity of Vinca herbacea on Human Ovarian Cancer Cell Line

Background: It seems that Vinca. herbacea has an anti-tumor effect. Here, the immunotherapeutic effect of this compound is assessed against human ovarian cancer (SKOV3) cells because of the high incidence of this tumor in women. Materials and Methods: The cytotoxic activity of V. herbacea extract against human ovarian cancer (SKOV3) cells was determined by MTT assay. The apoptosis-inducing pote...

Anti-angiogenic Effects of Metformin, an AMPK Activator, on Human Umbilical Vein Endothelial Cells and on Granulation Tissue in Rat

Objective(s)Metformin is well known for activation of AMP-activated protein kinase (AMPK). AMPK activation inhibits mammalian target of rapamycin (mTOR) as a key signaling process in cell proliferation. Recent epidemiological studies demonstrate that metformin lowers the risk for several types of cancer in diabetic patients. Concerning the critical role of angiogenesis in the incidence and prog...
